裙带菜水提液的免疫调节及对肿瘤细胞增殖作用的研究

Study of the lmmunoregulatory and Antitumor Activities of Undaria Pinnatifida Extract in Vitro

摘要: 背景与目的: 探讨裙带菜水溶性提取液(以下简称水提液)的体外免疫调节活性及抗肿瘤作用。 材料与方法: 采用3H-TdR掺入法分析4个不同浓度组(0.625、1.25、2.5和5 mg/ml)的裙带菜水提液对小鼠脾细胞的增殖作用;采用生物学方法测定小鼠单核细胞肿瘤坏死因子(Tumor necrosis factor-α, TNF-α)的产生。用MTT法测定裙带菜水提液对肿瘤细胞的抑制效应。 结果: 裙带菜水提液各浓度组都可显著刺激淋巴细胞的增殖;在体外能激活小鼠单核细胞并产生大量TNF-α,促进其吞噬或杀伤肿瘤细胞;裙带菜水提液对K562、S180、TE-13、SK-OV3 4种肿瘤细胞的体外增殖均有明显的抑制作用。 结论: 裙带菜水提液能显著刺激小鼠淋巴细胞的增殖,显著促进小鼠单核细胞的吞噬功能和细胞因子的产生;并对肿瘤细胞的生长具有明显的抑制作用。

Abstract: BACKGROUND & AIM: To investigate the immunoregulatory and antitumor activities of Undaria pinnatifida extract. MATERIAL AND METHODS: Effects of Undaria pinnatifida extract on proliferation of spleen cells of mice were analyzed by 3H-TdR incorporation methods, effects of Undaria pinnatifida extract on stimulating tumor necrosis factor -α (TNF-α) secretion from mice monocytes were examined by a biological method, inhibition of tumor cells by Undaria pinnatifida extract with MTT technique. RESULTS: All concentrations of Undaria pinnatifida extract had effects of promoting the proliferation of spleen cells of mice. In vitro, undaria pinnatifida extract could activate mice monocytes to produce more TNF-α, and inhibited proliferation of four types of tumor cells. CONCLUSION: The present study indicateed that Undaria pinnatifida extract had the effects of promoting the proliferation of mice spleen cells, and induced mice monocytes to produce TNF-α.and significantly inhibited the growth of four kinds of tumer cells.
